Highly divergent neuropeptide – non-coding RNA regulatory networks underpin variant host-finding behaviours in Steinernema species infective juveniles

چکیده

We conducted a transcriptomic and small RNA analysis of infective juveniles (IJs) from three behaviourally distinct Steinernema species. Substantial variation was found in the expression shared gene orthologues, revealing signatures that correlate with behavioural states. Ninety-seven percent predicted microRNAs are novel to each Surprisingly, our data provide evidence new family non-coding transcripts overlap neuropeptide loci, which influence microRNA regulation genes. These suggest differences expression, isoform variation, interactions could contribute within genus.

Location of the Phasmids on Infective Juveniles of Steinernema glaseri.

Scanning electron microscopy revealed the location of the phasmids on infective juveniles of Steinernema glaseri. The phasmids are located about 40% of the tail length posterior to the anus and are at or near the same level. Instead of being in the center of the lateral fields, they are located just ventral to the lateral fields, or interrupting the ventral-most lateral ridge. The phasmids were...

SOX2OT, a long non-coding RNA involved in autophagy regulation

Summary: SOX2 overlapping transcript (SOX2OT) is a long non-coding RNA associated with cancer pathogenesis. It contributes to a variety of cellular functions and recent evidence propounds its association with autophagy process. It has been showed that SOX2OT can regulate the expression of different autophagy associated factors in human cells with different mechanisms, however more remains to ...

Capturing protein-coding genes across highly divergent species.

DNA hybridization capture combined with next generation sequencing can be used to determine the sequences of hundreds of target genes across hundreds of individuals in a single experiment. However, the approach has thus far only been successfully applied to capture targets that are highly similar in sequence to the bait molecules. Here we introduce modifications that extend the reach of the met...

Intracellular Freezing in the Infective Juveniles of Steinernema feltiae: An Entomopathogenic Nematode

Taking advantage of their optical transparency, we clearly observed the third stage infective juveniles (IJs) of Steinernema feltiae freezing under a cryo-stage microscope. The IJs froze when the water surrounding them froze at -2°C and below. However, they avoid inoculative freezing at -1°C, suggesting cryoprotective dehydration. Freezing was evident as a sudden darkening and cessation of IJs'...

Simple In Vivo Production and Storage Methods for Steinernema carpocapsae Infective Juveniles.

Methods are described for standardized in vivo production, rapid harvest, and storage, in a concentrated form, of infective juveniles of the entomopathogenic nematode, Steinernema carpocapsae Mexican strain Kapow selection. Nematodes were stored in nematode wool configurations, consisting of mats of intertwined infective juveniles. Freshly harvested nematodes are readily available in adequate q...
